Role of integrin in the interaction between podocyte and glomerular basement membrane

Objectives: Podocyte cells and glomerular basement membrane (GBM) are main components of glomerular filtration barrier. We hypothesized that podocyte cells adapt to physical or chemical stimuli through changing in its interaction with GBM.  Methods: We investigated human kidney tissues obtained from the patients with trauma (control), membranous nephropathy (MN), and minimal change disease (MCD). We investigated the interaction between podocyte cells and GBM using antibodies for integrin (Itg) α3β1, Itg (αV)β3, UPAR, and fibronectin. Results: A total of 45 kidney tissues (control: 15; MN: 15; MCD: 15) were examined. The results showed that Itg α3β1 was ubiquitously expressed both in glomerulus and tubules, while Itg β3 was only in glomerulus (Figure 1). Control group showed a higher Itg β3 expression than that of MN and MCD (p< 0.0001), while had lower expressions of UPAR and fibronectin (p< 0.0001). There were no differences in Itg β3, UPAR, and fibronectin between MN and MCD groups (Figure 2). Conclusions: This study suggests that Itg αVβ3 has a specific role in the interaction between podocyte cells and GBM of glomerulus. In particular, we believe that Itg αVβ3 plays a important role in association with UPAR and fibronectin, in the glomerular diseases such as MN or MCD.
